To enter the Thieves Guild you must first travel to Riften and speak to Brynjolf to begin the test. He'll then tell you to meet him in the headquarters.To get to the headquarters, just drop down to the water-level platform that's below the merchant stalls in the center of the town. From there, you'll enter a door to the "Ratway". Travel through these sewers to come to the Ragged Flagon, a bar hidden beneath the city. From there, you just have to move through a tunnel on the backside of the bar and enter a door leading to the main part of the guild, the Cistern.I'm not quite sure if that was what you were asking.
